I’ll have a full review in the Mosen At large podcast when Spaces is released to everyone, in the same way that I did the 90 minute Clubhouse presentation. But the short answer is yes, it’s accessible. All buttons clearly labelled, and what’s great is that you can enable captions so Deaf or hearing impaired people aren’t excluded.
It is 100 percent accessible. I've been playign with it since march or so. and it sounds better than clubhouse. Picture the sound of facetime or duo but sounding clearer even than that.

I read that on IOS, everyone can already set up Spaces. How to do that? Or are Spaces not available for everyone even on IOS?
Twitter Spaces Availability
The best source for info on the Twitter Spaces availability is: https://twitter.com/TwitterSpaces?ref_src=twsrc%5Egoogle%7Ctwcamp%5Eserp%7Ctwgr%5Eauthor
The short version is that it will be increasingly available over the next few weeks.
