There are a good number of them - the Apple Maps speaks with voiceover, Google Maps do likewise, there is Blind Square which cost some money, there is Sound Scape by Microsoft which is free, and the latest to join is Good Maps.
I'll say this much, if you know and have heard of Mike May during the days of Arkinstone and his vision for accessible GPS technology, moving on to form the company Sendero, well, he is involved with Good Maps and it is working hard on indoor and outdoor navigation.
You will have to try all of these options and really determine what best fits your need.

what you do, is set markers at the the points on your route. Once the route is complete, then you use these markers to go from where are to where you want to go to on the route that you've already picked using the markers.
