the best mail VoiceOver voice in my opinion, is Alex. it is all so subjective though. I like Alex for reading books and other long material. the breathing this voice does is amazing! the Alex voice was so different in Mac OS X Leopard, but the refinements every year on the Mac side to this voice, it just keeps getting more like leopard every year.
As has been said previously, personal preference is what it boils down to. In my opinion the Vocalizer voices tend to be a bit lax on intonation and pitch, and unless you use the compact ones you're bound to get a noticeable delay. But having said that I like these and all the voices. I experienced constant speech crashes for awhile there, but that problem seems to have been resolved with this new internal file system which Apple has implemented on the Mac. Also props to Apple for the Alex voice, with his breathing.
I generally use Alex for VoiceOver, but I also use Tom for certain applications. I also used to use one of the British voices, though I don’t remember which one now, but that was for a very specific situation where having a “proper” English dialect was necessary.
I will echo other comments here in saying that the best voice for each individual will be different. I’m sure there are some VoiceOver users who like Fred, even though I could never use him for any length of time without getting a splitting headache.

The reason I say this, is because a bunch of you started saying the best VoiceOver voice is your choice. But the post said, what is the best VoiceOver voice you suggest. See the difference? Asking what's the best VoiceOver voice and what's the best VoiceOver voice sy suggest are two different ways of asking.
So, my answer is: I like Alex. Alex may be your best shot.

Best Voice for What Purpose?
Clearest in noisy situations? Samantha. Most natural? Alex. Most responsive but sounds horrible? Fred.
I wish there was a voice that combined those attributes: responsiveness, clarity, and naturalness. So far, Alex is the best compromise I can find.
