Yes, I have definitely seen this problem too - Not new to iOS 10.
Often when I arrow up and down through a note or mail message I am editing on my iPhone, VO will speak "mis-spelled" or say a fraction of a line. Arrowing up and then down past the offending line again seems to correct this as if the screen were reformatted.
Also, when I arrow to the top line or bottom line of an edit field, the first or last line will not be spoken (as if the line were blank) when hitting up arrow or down arrow to get to the first or last line. The only way I know the first or last line is there is to navigate by word and not by line with the up/down arrow keys.
Don't know why it works this way or can't be fixed. VO is not perfect.
